چکیده
Cytotoxic necrotizing factor (CNF) toxins, isolated from certain Escherichia coli strains known t o cause intestinal and extra intestinal infections, induce reorganization of the actin cytoskeleton and generate hyperploidy in adherent cell lines. We have examined the effect of CNF toxin on one of the few cell types that naturally increase nuclear DNA content, megakaryocytes. Our studies show that only hematopoietic cells capable of differentiating along the megakaryocyte lineage responded t o the CNFZ toxin by becoming polyploid and by reorganizing actin. The K562, HEL, and CHRF-288-11 cell lines can be induced with phorbol ester t o differentiate along the megakaryocyte lineage, and these cells also respond t o the toxin with increased DNA content and actin cytoskeletal rearrangements. Interestingly, treatment of the K562 and HEL cell lines with CNF2 does not result in an increase in production of the megakaryocytic marker glycoprotein Illa, unlike phorbol ester treatment. Conversely, two

متن کاملA Pilot Study of Nuclear Instability in Archived Renal and Upper Urinary Tract Tumours with Putative Ochratoxin Aetiology
DNA ploidy measurement has been applied uniquely to wax-embedded tissue of primary renal cell and metastatic tumours of a key experimental researcher on porcine ochratoxicosis, a control, and four transitional cell carcinomas from cases of Balkan endemic nephropathy. Primary renal tumour was diploid, and hyperdiploid metastasis was within the lower ploidy range for typical renal cell carcinoma....
متن کاملGains from diversification on convex combinations: A majorization and stochastic dominance approach
By incorporating both majorization theory and stochastic dominance theory, this paper presents a general theory and a unifying framework for determining the diversification preferences of risk-averse investors and conditions under which they would unanimously judge a particular asset to be superior. In particular, we develop a theory for comparing the preferences of different convex combination...
متن کاملImproved immunogenicity of tetanus toxoid by Brucella abortus S19 LPS adjuvant.
BACKGROUND Adjuvants are used to increase the immunogenicity of new generation vaccines, especially those based on recombinant proteins. Despite immunostimulatory properties, the use of bacterial lipopolysaccharide (LPS) as an adjuvant has been hampered due to its toxicity and pyrogenicity. Brucella abortus LPS is less toxic and has no pyrogenic properties compared to LPS from other gram negati...
متن کاملSteady electrodiffusion in hydrogel-colloid composites: macroscale properties from microscale electrokinetics.
A rigorous microscale electrokinetic model for hydrogel-colloid composites is adopted to compute macroscale profiles of electrolyte concentration, electrostatic potential, and hydrostatic pressure across membranes that separate electrolytes with different concentrations. The membranes are uncharged polymeric hydrogels in which charged spherical colloidal particles are immobilized and randomly d...
